A lox P site was inserted 5' of exon 2 and a floxed PGK-neomycin selection cassette was inserted 3' of exon 4. Transient cre expression in these ES cells resulted in excision of the Neomycin selection cassette as well as exons 2, 3, and 4. Northern blot analysis indicated the presence of a truncated mRNA. However, no protein product was detectable by Western blot analysis. (J:123295)
