A lox P site was inserted 5' of exon 2 and a floxed PGK-neomycin selection cassette was inserted 3' of exon 4. Transient cre expression in these ES cells resulted in excision of the neomycin selection cassette and left exons 2, 3 and 4 flanked by loxP sites. (J:123295)
