A targeting vector was designed to flank the sonic hedgehog processing domain in exon 3 with loxP sites. The second loxP site was followed by a Myc epitope tag with a stop codon and an FRT site flanked neomycin resistance gene cassette downstream of the stop codon. The neo cassette was removed by crossing with mice expressing flp recombinase. (J:109101)
