A targeting vector was designed to replace exon 6 and part of exons 5 and 7 with a neomycin resistance gene. A termination was introduced into exon 5 in the pro-protein domain as a result, and any protein synthesized from this allele would be truncated and non-functional. Absence of protein in mutant cerebellum was confirmed by Western blot analysis. (J:107127)
