LoxP sites were inserted to flank exons encoding the DSL (Delta-Serrate-Lag2) domain required for binding to Notch. Expression of cre recombinase is expected to knock out gene function by removing exons 4 and 5 (the DSL encoding exons) and causing a frame shift that generates a stop codon after the start of exon 6.
