The last 426 amino acids of the locus were replaced with the corresponding region of Notch1 and a neomycin resistance cassette by homologous recombination. The resulting allele consists of the first 2044 amino acids of the endogenous locus fused to the last 445 amino acits and 3' UTR of Notch1. RT-PCR confirmed correct recombination of the locus in mutants. (J:107000)
