The transgene constructs encode a T cell receptor (TCR) that recognizes the Ld class I major histocompatibility complex (MHC) antigen SIYRYYGL peptide in the context of the H2b MHC class I molecule. Rearranged genomic cosmids containing the entire Tcra and Tcrb coding and promoter regions from the cytotoxic T cell clone 2C were isolated and coinjected into fertilized eggs. Transgenic H2b mice have large numbers of T cells in the periphery that express the transgenic TCR. In the periphery of transgenic H2d mice, T cells expressing the transgenic TCR are deleted.
