The rat phosphoenolpyruvate carboxykinase 1, cytosolic promoter was used to drive expression of cre recombinase in the renal proximal tubule. The promoter was mutated in such a way that expression in the liver was reduced by 60% and increased in the kidney by 10-fold in mutant mice. The construct was inserted into ES cells lacking Hprt.
