A 2200 bp mouse genomic DNA fragment containing the S100 calcium binding protein A4 (S100a4) gene sequence, including the second intron and followed by an SV40 polyadenylation signal, was placed under transcriptional control of the long terminal repeat (LTR) of mouse mammary tumor virus (MMTV). Line 507 was shown by Southern blot analysis to have integrated 2-4 copies at a single site, apparently with "a minor rearrangement." Northern blot analysis demonstrates high levels of transgene-derived mRNA in lactating mammary glands of transgenic mice; the transcript also is present in thymus and spleen and at a low level in salivary gland, kidney and lung. S100A4 protein is detectable by immunoblot analysis in lactating mammary glands of transgenic mice, but not of their wild-type littermates. (J:36642)
