A transgene containing an expression construct for yeast cAMP phosphodiesterase (SRA5) under the control of the human insulin promoter randomly inserted into the Prkdc gene, disrupting the normal function of this gene. Expression of Prkdc was shown to be abolished by RT-PCR analysis on RNA derived from homozygous spleen and thymus. Expression of the transgene was negligible. Complementation analysis with Prkdcscid mice confirmed that the insertion mutation is an allele of the Prkdc gene. (J:44390)
