The transgene comprises nucleotides -421 through +28 of the rat probasin gene, containing the promoter, upstream of a cDNA encoding hemagglutinin (HA) epitope tagged, myristoylated human AKT1, the product of the human ortholog of mouse thymoma viral proto-oncogene 1. RT-PCR and immunoblot analysis with antibody to the HA tag revealed prostate-specific expression of both mRNA and protein; immunoblot analysis of protein extracts of individual prostatic lobes and other tissues from the urogenital tract localized expression to the ventral prostate and, at low levels, to the lateral prostate.
