This transgene consists of the mouse alpha myosin heavy chain (Myh6) promoter and a truncated Pik3ca (p110) mutant that has Pik3rl (p85) binding domains but lacks the kinase domain. Myh6 drives the expression of the dominant-negative Pik3ca in cardiac myocytes. Immunoprecipitation and western blotting confirmed transgene expression exclusively in the heart and a lipid kinase indicated that phosphatidylinositol 3-kinase activity was decreased by 77%.
