Two-step homologous recombination was used to delete 70 of the Tcrb locus (475 kb). The first targeting event replaced Dbeta1-Jbeta1 cluster with a loxP flanked neo and subsequent cre expression in the ES cell removed neo leaving a single loxP site. The second targeting event inserted another loxP flanked neo gene into the Vbeta1 exon. Subsequent cre expression in the selected ES clone removed large 475 kb region including the 3' half of the 5'Vbeta region, a central large intron of the locus and the first Dbeta-Jbeta cluster.
