A targeting construct was designed to replace the T-cell receptor gamma variable 1 gene as well as the joining region and exons 1 and 2 of the T-cell receptor gamma constant 4 gene with a neomycin resistance cassette. Removal of this region would prevent any rearrangement and the possibility of pairing other variable gamma genes with an otherwise intact constant 4 gene. Absence of Tcrg-V1 mRNA in RT-PCR of mutant spleen and thymus confirmed successful ablation. (J:105477)
