Cre mediated recombination removed a floxed neo, leaving exon 1, containing the ATG, replaced by a c-Myc epitope tagged Epas1 cDNA such that knock-in expression depended upon the endogenous regulatory sequence. Mutant ES cells did not express Hif1a protein, as shown by Western blot and EMSA analyses. Immunoprecipitation and immunoblot detected expression of the knock-in sequence. (J:97164)
