A targeting vector was designed to flank exons 15-21, which encodes 40% of domain V, consisting of laminin EGF-like repeats. Upon removal of the sequence, splicing from exon 14 to exon 22 should result in an out of frame transcript. The neo cassette used for selection purposes was removed by flp mediated recombination.
