Cre recombinase was used to invert a 75-kb loxP-flanked region of the locus. The region encompasses all of the J-alpha gene segments, the C-alpha gene segment and enhancer E-alpha. In this allele the J-alpha gene segments lie in the opposite transcriptional orientation compared to the V-alpha gene segments. Accordingly, initial V-alpha to J-alpha rearrangements on the allele occur by inversion, resulting in retention of the Tcrd genes that lie within the locus in the mature alpha-beta T cells.
