A bacterial lacZ gene was inserted into the gene such that the endogenous gene promoter drives expression of beta-galactosidase. RT-PCR analysis detects a high level of gene transcript in the whole brain, subcortical region, brainstem and spinal cord, moderate levels in cortex, cerebellum, olfactory bulb, eye, testis and ovary and low levels in cortex, cerebellum, olfactory bulb, eye, Harderian glands, lung, liver, pancreas, kidney, spleen, thymus, lymph nodes, skin, gallbladder, urinary bladder, pituitary gland, adrenal gland, salivary gland, tongue, stomach, small intestine, colon, coagulating gland, prostate gland, uterus and white fat. Beta-galactosidase expression from this allele is detectable in brain, spinal cord, eyes, skin, and testis. Moderate to strong lacZ expression was detected in cells scattered throughout the brain and spinal cord. In the brain, most striking expression was detected in white matter, thalamus and brainstem. In the spinal cord, gray matter showed moderate, scattered staining while only a few cells in the white matter showed weak staining. Strong expression was detected in the ganglion cell and inner nuclear layers of the retina. In skin, strong staining was observed in a few cells surrounding hair follicles. Strong expression was also detected in the seminiferous tubules of the testis.
