A bacterial lacZ gene was inserted into the gene such that the endogenous gene promoter drives expression of beta-galactosidase. RT-PCR analysis detects a high level of gene transcript in the brain cortex, moderate levels in whole brain, subcortical region, cerebellum and olfactory bulb, and low levels in brainstem, spinal cord, eye, Harderian glands, heart, lung, liver, pancreas, kidney, spleen, thymus, lymph nodes, skin, gallbladder, urinary bladder, pituitary gland, adrenal gland, salivary gland, skeletal muscle, tongue, stomach, small intestine, cecum, colon, testis, epididymis, seminal vesicle, coagulating gland, prostate gland, ovary, uterus and white fat. However, beta-galactosidase expression from this allele is detectable in only in brain, kidney and testis, with moderate lacZ expression detected in some regions of the whole mount brain including areas of the cortex and hippocampus. On coronal sections of brain, very weak staining was observed in a few cells of the pyramidal cell layer in hippocampus. Strong, scattered expression was observed in renal medulla and papilla. Strong staining was also observed in a few spermatogenic cells in the testis.
