The targeting vector was designed to replace the coding sequences from exon 2, including the translation initiation codon, the N-terminal half LIM motif, part of the LIM1 domain, and 0.5 kb of intron 2 with a lacZ reporter gene and a loxP flanked neomycin resistance cassette. Transcription of the lacZ insertion was evident in various stages of embryonic development. Absence of endogenous transcript and protein in mutants was confirmed by Northern and Western blots, respectively. (J:100289)
