A PGK-neomycin resistance gene in reverse orientation replaced exon 7 in order to abolish the core region of the homeodomain. A SIGP cassette was inserted downstream of the neo, thereby inserting a splicing acceptor, IRES, GFP and polyA signal. The expressed transcript would be the fusion of exons 1-6 of Lhx8 and the IRES-GFP-polyA. eGFP expression was not detected by fluorescent microscopy and immunohistochemistry, but was detected by in situ hybridization, suggesting that the translation efficiency of eGFP is very low or it may be suppressed in the mice for an unknon reason.
