A gene trap vector containing beta-galactosidase and the engrailed 2 promoter integrated into intron 28 at amino acid 1295. The mutant allele was predicted to express endogenous amino acids 1-1295 fused to Beta-gal. Mutant tissues analyzed by Northern blot did not express transcript corresponding to the 3' UTR probe. LacZ signals were detected. Shorter transcripts were detected, possibly resulting from alternative splice forms. The testis splice form, which normally contains an extra 251 base pairs encoding a unique 30 amino acid N-terminus, only contained 204 residues of the wild-type sequence. The 3.5 kb kidney transcript begins downstream of the insertion site, and remained unaffected by the gene trap insertion. (J:102137)
