The transgene was created by targeted insertion of a "humanized" green fluorescent protein (GFP) coding sequence into the bacterial artificial chromosome (BAC) RP23-88K7. The GFP sequence integrated at position +15 of Ren1, which is centered within the approximately 250-kb BAC, with deletion of exon 1 and approximately 200 base pairs of intron 1. Combined SKY-FISH analysis localized the integration site to Chromosome 3. GFP is expressed in adrenal glands of transgenic fetuses from about day E13 and in fetal renal arteries from about E14 through birth; it is also expressed in fetal adrenal and gonadal arteries. After birth, expression in renal arteries becomes progressively restricted, so that only juxtaglomerular cells fluoresce by approximately 12 weeks of age. Larger adult renal arteries can be induced to resume GFP expression by treatment with the ACE inhibitor captopril. Low levels of GFP expression occur in submandibular glands of male transgenic mice.
