The modified estrogen receptor hormone-binding domain ERT2 was inserted in-frame with the coding region for the Meis1a isoform using the ACN cassette, which excises itself following passage through spermatogenesis. Insertion of the vector eliminated the Meis1b coding region. The mutant allele is predicted to encode an inactive protein unless tamoxifen is present. In mutants, a protein band for the expected size of Meis1a-ERT2 was present, though at a much lower level than wild-type protein. (J:98307)
