A targeting vector was designed to replace serine 21, located on exon 1, with alanine (S21A). A floxed neomycin resistance cassette was inserted at the 3' end of exon 1. The neomycin cassette was subsequently excised from properly targeted ES cells by transient transfection with a cre expressing plasmid. (J:98263)
