The 7 exons encoding the alphaLm globular (LG) 5 region were removed by fusion of the DraIII and SmaI restriction sites, creating an in-frame deletion. A neo-HSV-tk flanked by loxP sites was inserted downstream of the stop codon and polyA tail. The last 341 residues of the C terminus were deleted and the linker region between alpha1LG3 and alpha1LG4 and a short sequence of the first Beta-strand of the alpha1LG4 domain were left intact.
