A loxP-flanked neo replaced exons 4-10 (G321 to A626), which encode the C-terminal half of the protein that includes the RING finger domain. Northern blot showed the presence of three new transcripts. The longest form putatively corresponds to a partially spliced mRNA stabilized by the polyA signal of neo. The shortest form was suggested to be created by splicing of the donor site of exon 3 to a cryptic acceptor site in the neo, which created an in-frame stop codon 7 amino acids downstream. It could not be determined if a truncated protein without the RING finger domain was expressed. (J:95986)
