A 2.3 kb fragment containing the mouse Alb promoter/enhancer elements was cloned upstream of a mouse Trp53 minigene containing an Arg to Ser substitution at codon 246 (corresponding to human codon 249). This transgene was not linked with the wildtype Trp53 allele, and was incorporated in a chromosome other than the one carrying endogenous Trp53. Transgene incorporation was confirmed by Southern blot, and expression was confirmed by RT-PCR. (J:47088)
