A targeting vector was designed to replace exons 1-4, which contain most of the coding sequence, with a neo gene lacking the polyadenlylation sequence. RT-PCR and sequencing revealed that exons 5 and 6 were transcribed after the stop codon of the neo mRNA, as part of the 3' UTR. Absence of protein from exons 5 and 6 was further confirmed by Western blot analysis of kidneys from mutant animals. (J:95282)
