A loxP site was inserted upstream of exon 2 and a floxed PGK-neomycin cassette was inserted downstream. Transient transfection of properly targeted ES cells with a Cre-expressing plasmid resulted in excision of the selection cassette together with exon 2. Immunoblot analysis confirmed the absence of protein product. (J:94756)
