Exon 1, which contains the initiation codon and sequences encoding the signal peptide, was replaced with a loxP-flanked neomycin selection cassette. The selection cassette was removed through Cre-mediated recombination, leaving a single loxP site in place of exon 1. RT-PCR and in situ hybridization analysis confirmed that no detectable transcript is expressed from this allele. (J:126239)
