A loxP site flanked neomycin resistance gene cassette replaced a 1.9 kb section containing DNase I hypersensitivity site IV (HS IV), an Il4 silencer located between Il4 and Kif3a. Transient Cre recombinase expression removed the neo cassette in ES cells. DNase hypersensitivity analysis of mutant cells showed a loss of HS IV, but no disruption of the other hypersensitivity sites. (J:94039)
