A targeting vector was inserted into the 3' region of the gene, resulting in loxP sites flanking exons 8-10 as well as an inserted polyA/stop signal from the C2 gene and exons 2 and 3 of the Foxo1 gene (with an IRES-eYFP cassette in exon 3). Crossing with FLP expressing mice excised an FRT flanked neo that was also inserted downstream. The mutation allowed normal functioning until activation of Cre recombinase. Cre causes the fusion of exons 1-7 of Pax3 to a region of Foxo1 containing exons 2 and 3, as well as 6.5 kb of the UTR and genomic sequence. Simultaneously, an IRES-eYFP marker is expressed. (J:93444)
