A targeting vector was designed to introduce a p.S389A mutation (T>G) in exon 11. This should abolish phosphorylation of the protein at that amino acid residue in the encoded peptide. A loxP site flanked neomycin resistance gene cassette was introduced downstream of exon 11 and subsequently excised by crossing with Cre expressing deleter mice. Expression of the mutated allele was confirmed by RT-PCR analysis. (J:93328)
