A targeting vector was designed to replace exons 22 through 25 with a neomycin resistance gene, stop codons in all three reading frames, and an IRES-Beta-galactosidase cassette. The deleted sequence encoded the 7 transmembrane domain. Northern blot analysis of mutants indicated a lack of transcript for this domain. Two short, weakly expressed transcripts were present in mutants, one corresponding to exons 1-20 and the other corresponding to the truncated mRNA fused to Beta-gal. Ablation of wild-type protein was confirmed by Western blot of epididymal membrane samples, and lacZ activity was demonstrated by BLUO-GAL staining. (J:93019)
