The complete coding region was replaced with a loxP site following Cre-mediated excision of a floxed sequence including a neo and the endogenous coding sequence. RT-PCR and Western blot confirmed that the gene was abolished by demonstrating lack of transcript and protein, respectively. (J:92791)
