Transgenic mice carry two coinjected constructs containing rearranged mouse TCR alpha (Tcra-V2, Tcra-J(alpha26)) and TCR beta (Tcrb-V5, Tcrb-D(beta2), Tcrb-J(beta2.6)) chains isolated from T cell clone 149.42. This transgenic T cell receptor recognizes ovalbumin residues 257-264 (SIINFEKL) in the context of H2Kb. The rearranged Tcra (alpha chain) cDNA specific for ovalbumin was isolated from T cells (clone AD10) and is expressed under the control of a 2-kb fragment of the H-2Kb promoter, a 700-bp fragment including the Ig H chain enhancer located 5' to the promoter, and a 1.7-kb BamHI-PstI fragment containing the 3' half of the human adult 8-globin gene (including part of exon 2, the exon 2-exon 3 C.F6 TCR a-chain gene that was truncated at the 3' XbaI site intron, exon 3) and polyadenylation signal. The Tcrb (beta chain) construct consists of a genomic fragment containing the Tcrb coding and endogenous promoter regions from a T cell clone (B3) specific for ovalbumin. (J:92867)
