The 5' UTR and the first 208 codons of exon 1, encoding the N-terminal half of the forkhead domain, were replaced with a targeting vector containing a floxed PGK-neo and FOXO1a cDNA, encoding a constitutively active form of the protein. RT-PCR and Western blot showed lack of wild-type transcript and protein, respectively, in mutant embryos. (J:92247)
