A targeting vector was designed to replace exons 7-9, which encode residues 180-255, with a splice acceptor and IRES-Beta-neo-pA cassette, and thereby disrupt the second DNA helicase domain. Southern blot confirmed recombination in ES cells, and Northern blot of mutant embryos showed no transcript containing the 3' end of the gene. LacZ expression was as expected in mutant embryos.
