A loxP site was inserted 560 bp upstream of the ATG for the pIII transcript and another loxP site was inserted 90 bp downstream of the first exon encoding the pIV transcript, followed by an FRT-flanked neo. Transient FLP recombinase expression excised the neo and mice generated were crossed with CMV-Cre mice to excise the floxed pIII and pIV sequence. RT-PCR indicated transcripts of both were undetectable in mutant tissues. Gene expression from pI was unaffected. (J:92170)
