Part of the first exon and first intron was replaced by a lacZ-neomycin fusion gene, such that beta-galactosidase should be expressed under the control of the endogenous promoter. Northern and western blot analysis demonstrated an absence of encoded mRNA and protein, respectively, in homozygous mice. (J:55800)
