Exon 2 was replaced with an IRES and floxed PGK-neo cassette. Southern blot confirmed recombination and RT-PCR indicated no mRNA in mutant pups. Immunoblot demonstrated lack of expression of the full-length protein in fetal liver mast cells. (J:91790)
