An expression cassette of the hypoxanthine phosphoriboyltrasferase (HPRT) gene replaced the promoter region, and sequences encoding the transcription activation domain and part of the DNA binding domain of the gene. Northern and RT-PCR analysis showed that a null allele was obtained. (J:51402)
