A targeting vector containing translation stop codons, the promotorless beta-galactosidase gene, and MC1 promoter-linked neo resistance gene replaced most of the coding region (amino acids 27-455). Southern blot and PCR confirmed recombination, however, heterozygous embryos did not express beta-galactosidase activity. (J:91247)
