A neo cassette replaced exons 7 and 8, which encode part of the kinase domain. Sequencing of mRNA from the mutant allele confirmed lack of exons 7 and 8 and indicated a premature stop codon was created, thereby generating a truncated protein that contained only a part of the kinase domain. A transfection assay showed that mutant protein did not function as the wild-type. (J:91363)
