The first 654 bp of the single exon gene was replaced with a lacZ reporter and a neomycin resistance gene, with the initiation codon of lacZ inserted in frame with original the transcription start site. Southern blot confirmed recombination and reverse transcription PCR demonstrated absence of normal mRNA and presence of lacZ mRNA in mutants. (J:89708)
