Exon 1, encoding the start codon and signal peptide, and a portion of exon 2, encoding the IgV domain, were replaced with a neo cassette inserted by homologous recombination. Protein was undetected on the surface of homozygous mutant CD4+ T cells via flow cytometric analysis. Western blot analysis further confirmed the absence of normal protein in homozygous mutant mice.
