A modified mouse Dll1 cDNA encoding a truncated protein, lacking the majority of the intracellular domain, was adjoined to 1.2 kb of the mouse Mesp2 promoter. The construct also contained an SV40 3' UTR and polyA signal. The Mesp2 promoter was expected to drive expression of the mutant Dll1 cDNA in the anterior presomitic mesoderm corresponding approximately to somitomere S-I. The mutant protein is expected to lack 141 C-terminal residues, leaving only 12 intracellular residues adjacent to the transmembrane domain. (J:88580)
