Insertion of a translational stop codon at the end of the POU homeodomain encoding exon by homologous recombination results a truncated protein. All sequences C-terminal to the DNA-binding domain are removed from proteins translated from this allele. Western blots demonstrated that this truncated protein is produced and stable in vitro.
