Homologous recombination was used to replace a 3.2 kb genomic fragment including the exon encoding the second immunoglobulin domain with a neomycin cassette in antisense orientation relative to transcription. RT-PCR and flow cytometric analysis indicated an absence of transcript and protein, respectively, in homozygous mutant MEFs. (J:84973)
